What are the effects of taking 500 mg of metformin twice per day and also having three to four glasses of wine with supper?

Answer

Well, most likely you'd get drunk. There's also a reasonable possibility of that amount of alcohol affecting your blood sugar. See alcohol and diabetes for more information.

Alcohol blocks the liver's ability to release stored sugar. That amount might lead to hypoglycemia.
